T., Hemmerlé, J., Gullberg, D., Kedinger, M., Sorokin, L., Orend, G., Simon-Assmann, P. “The laminin response in inflammatory bowel disease: protection or malignancy?” 2014. PLoS One 9(10):e111336. http://www.ncbi.nlm.nih.gov/pubmed/25347196 A recent report from Spenlé and colleagues published in PLoSOne describes abnormal overexpression of lamininα1 andα5 in human andmouse colitis samples that was associated with p53 expression. To further explore the relevance of these observations, the authors utilized transgenic mice that overexpress intestine-specific Lama1 or Lama5. Thesemicewere less vulnerable to inflammation butmore prone to cancer progression in vivo (Fig. 1). These results define unique laminin function related to gastrointestinal inflammation and cancer. 2). The matrix corroded: podosomes and invadopodia in extracellular matrix degradation.
Podosomes and invadopodia are unique actin-rich adhesions that establish close contact to the substratum but can also degrade components of the extracellular matrix. Accordingly, matrix degradation localized at podosomes or invadopodia is thought to contribute to cellular invasiveness in physiological and pathological situations. Cell types that form podosomes include monocytic, endothelial and...
متن کاملIntermediate filaments and the regulation of focal adhesion.
Focal adhesions are localized actin filament-anchoring signalling centres at the cell-extracellular matrix interface. The currently emerging view is that they fulfil an all-embracing coordinating function for the entire cytoskeleton. This review highlights the tight relationship between focal adhesions and the intermediate filament cytoskeleton. We summarize the accumulating evidence for direct...
متن کاملThe Emerging Role of PEDF in Stem Cell Biology
Encoded by a single gene, PEDF is a 50 kDa glycoprotein that is highly conserved and is widely expressed among many tissues. Most secreted PEDF deposits within the extracellular matrix, with cell-type-specific functions. While traditionally PEDF is known as a strong antiangiogenic factor, more recently, as this paper highlights, PEDF has been linked with stem cell biology, and there is now accu...
متن کاملMesenchymal stem cells can survive on the extracellular matrix-derived decellularized bovine articular cartilage scaffold
Objective (s): The scarcity of articular cartilage defect to repair due to absence of blood vessels and tissue engineering is one of the promising approaches for cartilage regeneration. The objective of this study was to prepare an extracellular matrix derived decellularized bovine articular cartilage scaffold and investigate its interactions with seeded rat bone marrow mesenchymal stem cells (...
متن کاملCombinatorial Scientific Computing: The Enabling Power of Discrete Algorithms in Computational Science
Combinatorial algorithms have long played a crucial, albeit under-recognized role in scientific computing. This impact ranges well beyond the familiar applications of graph algorithms in sparse matrices to include mesh generation, optimization, computational biology and chemistry, data analysis and parallelization. Trends in science and in computing suggest strongly that the importance of discr...
